Establishment of caprine airway epithelial cells grown in an air-liquid interface system to study caprine respiratory viruses and bacteria

Respiratory diseases negatively impact the global goat industry, but are understudied. There is a shortage of established and biological relevant in vitro or ex vivo assays to study caprine respiratory infections. Here, we describe establishment an system based on well-differentiated airway epithelial cell (AEC) cultures grown under air liquid interface conditions as experimental platform pathogens. The functional differentiation AEC was monitored confirmed by light immunofluorescence microscopy, scanning electron microscopy examination histological sections. We validated functionality studying Influenza D Virus (IDV) infection Mycoplasma mycoides subsp. capri (Mmc) colonization over 5 days, including monitoring infectious agents titration qPCR well colour changing units, respectively. inoculation with IDV showed that efficient viral replication takes place, revealed has marked tropism for ciliated cells. Furthermore, were successfully infected Mmc using multiplicity 0.1 several days. Altogether, these results demonstrate our newly-established can be used investigate host-pathogen interactions
